Glutathione Affinity Superflow 4

Glutathione Affinity Superflow 4 is manufactured by the optimized immobilization of high-grade glutathione to Sterogene`s trademarked Superflow 4 matrix. Glutathione/GST purification has proven to be a simple, effective method of purifying recombinant proteins expressed in the pGEX vector. Once bound to the resin, the GST-tagged protein can be eluted with glutathione, or it can be cleaved from the protein while still on the column. This results in the elution of the GST-free recombinant protein. 

This media provides an effective and efficient one-step purification for a variety of different expression systems. Along with the high binding capacity for GST-tagged proteins, Glutathione Affinity Superflow 4 shows low non-specific binding and is comparable in performance to the leading immobilized glutathione resins available.

Heparin Superflow 4

Heparin is a glycosaminoglycan carbohydrate and is a polymer of repeating units of disaccharides mostly composed of 2-O-sulfated iduronic acid and 6-O-sulfated, N-sulfated glucosamine. The average molecular weight ranges from 12-15 kDa. Heparins have multiple reactive groups; each repeating unit has one carboxyl group, one or more primary/secondary hydroxyl groups, and 2-2.5 sulfo groups.

Heparin Superflow 4 is used to selectively purify heparin-binding proteins in a single-step affinity or as a cation-exchanger in the purification process. It can be used successfully to purify plasma coagulation proteins, growth factors, lipoproteins, DNA/RNA binding proteins (DNA and RNA polymerases), serine proteases inhibitors, hormone receptors, extracellular matrix proteins (fibronectin, collagens), and certain enzymes (lipases). Heparin resin can also be used to purify exosomes from various body fluids and cell cultures. Heparins have a very high affinity for lentiviruses, and the heparin affinity resin can be specifically used to purify lentiviruses and lentivirus-based VLPs used in cell/gene therapy and vaccines.

Protein A Superflow 4

Protein A is a well-characterized antibody-binding protein composed of five homologous immunoglobulin-binding domains. Protein A binds strongly within the Fc region of several immunoglobulins and weakly to the Fab region of the VH3 class of immunoglobulins. 
Protein A resins are the most frequently used affinity chromatographic media and are a standard technique for the production of monoclonal antibodies. Sterogene’s Protein A Superflow 4 resin is alkali stable, has a high static binding capacity (50 mg/mL), has a high flow rate, and can be used multiple times without losing activity.

Protein G Superflow 4

Protein G is an immunoglobulin-binding protein. Compared to protein A, protein G binds to a broader range of immunoglobulins and binds to all the subclasses of IgGs from humans, mice, and rats. Sterogene’s Protein G-Superflow 4 resin is highly stable, with a high binding capacity (35 mg of human IgG/ml), and increased flow rate. This media can be regenerated and used multiple times.

Protein L Superflow 4

Protein L is a unique IgG-binding protein that binds to the kappa chain (kappa I, III, and IV of humans and kappa I of mice) of the light chain without disturbing the antigen-antibody interactions. Protein L binds to all the classes of antibodies such as: IgG, IgA, IgD, IgM, IgE, Fabs, and single-chain variable domains (scFv).

Sterogene’s protein L-Superflow 4 uses a recombinant protein L with five IgG binding domains. Protein L-Superflow 4 can be used to purify human IgGs, Fabs, or scFv from serum or any expression systems.

Aminogel

Aminogel effectively couples peptides and small ligands. This product is recommended primarily for the linkage of small ligands in which long hydrophilic spacer arms are needed to provide access of the protein to the binding moieties. Epoxy Activated Superflow 4

Epoxy Activated Superflow 4 is for stable immobilization of proteins through thiol, amino, or hydroxyl groups. Multiple functionaries allow more than one type of ligand immobilization.

Utilizing Sterogene`s patented Superflow 4 matrix, Epoxy Activated Superflow 4 exhibits significant increases in overall flow rates (>30 ml/min) and substitution levels (> 20 mg protein per ml resin).

Epoxy-Activated Superflow 4 features epoxy functional groups that react with nucleophilic groups on the ligand. During coupling, the epoxy group is activated by adjusting the pH.

Coupling is pH-dependent:
-For coupling thiol groups, pH 7-8 (forms a thioether linkage)
-For coupling amino groups, pH 8-10 (forms as a stable secondary amine linkage)
-For coupling hydroxyl groups, pH 12 (forms an ether linkage)

NHS-Activated Superflow 4

Sterogene’s NHS (N-hydroxysuccinimide) Activated Superflow 4 resin is a highly crosslinked agarose bead that has been pre-activated with an NHS-functional group. It is manufactured for the purpose of covalently coupling proteins, peptides, and antibodies through their primary amines in an easy single-step coupling reaction (Coupling efficiency > 80%).

The NHS-Activated Superflow 4 media reacts with the primary amine(s) on the ligand(s) to make a stable and covalent amide linkage. The coupled resin is highly stable with negligible leachability and can be used multiple times. The resin provides a long spacer arm and can be beneficial for making affinity chromatography media with small proteins or single-domain antibodies.

Buffers and Solutions

ALD Coupling Solution

ALD Coupling solution is used for the reduction of Schiff bases. Many aldehyde-activated resins require a reducing agent to form a stable secondary amine linkage from the temporary Schiff-base formed between the aldehyde and amine-containing protein.

Sterogene has developed a solution utilizing the milder reducing agent sodium cyanoborohydride (NaBH3CN). This material is sufficiently strong enough to couple the ligand to an activated aldehyde without reducing it. The ALD Coupling Solution is prepared using a highly purified form of the NaBH3CN solid that is free of NaBH4 contamination. This is desired amongst customers as increased levels of NaBH4 can cause a reduction of proteins. Please see Actigel ALD for further information.

Endotoxin Removal

ActiClean Etox

ActiClean Etox efficiently removes endotoxins from water, buffers, cell culture media, and protein solutions, including plasma proteins: Albumin, factor VIII, factor IX, IVIG with minimal non-specific binding. The immobilized ligand is non-toxic, and leaching is negligible. ActiClean Etox is easily regenerated for reuse. There is a Drug Master File available with the FDA. DNA Etox

DNA Etox is a high-efficiency, solid-phase reagent with a high affinity for endotoxin. Removing endotoxin from DNA is often difficult due to the similarity in the physicochemical characteristics of DNA and endotoxin. The most commonly used methods have limited utility in this area. Consequently, endotoxin removal is typically tedious and often incurs significant losses of DNA during the process. DNA Etox has high DNA recovery and can be used either in batch mode or in standard chromatography columns. DNA Etox can be readily regenerated with 1M NaOH. DNA Etox can also be sterilized by autoclaving.

Reduct Etox

ReductEtox resin is specifically designed for easy and rapid cleaning of endotoxins from proteins, water, buffers, and tissue cultures. ReductEtox can be easily sanitized and regenerated with 1M NaOH for multiple usages without loss of binding capacity. It can remove >99% of the endotoxin with superior protein recovery (~ 90%). The hydrophilic agarose support reduces nonspecific binding that normally occurs with less hydrophilic supports such as methacrylates or acrylamides. This results in higher binding capacity and protein yields.

Gel Filtration Resins, Activated Resins

Superflow 4

Superflow 4 beads exhibit the same high biological stability as the Uniflow media. The 4% beads are further stabilized by a chemical hardening reaction that enables larger porosity than the regular crosslinked 6% beads. Superflow 4 is an excellent choice as a support for immobilization or for gel filtration.

The media are stable in organic solvents and aqueous solutions in the pH range 2-14. Oxidizing materials should be avoided since they can cause partial decomposition of the polysaccharide chains. The media are also resistant to high concentrations of chaotropic agents, such as 4M KI or 4M NaSCN. They can be sterilized by autoclaving at 121 °C and pH 7 for 30 min.

Uniflow, Superflow and Ultraflow are provided as aqueous suspensions with 20% ethanol as preservative. They can be stored for years at 2-8°C. The media should not be frozen.

For packing, prepare a 50% slurry and de-gas it if necessary. Packing can be performed by gravity or pump. Make sure that the operating flow rate does not exceed the packing flow rate. Equilibrate the column with 2-3 bed volumes of buffer.

For elution, a peristaltic pump is recommended, as a consistent flow rate will aid in reproducible separations. The media can be cleaned in situ with one column volume of 0.5 M NaOH, which cleans and sanitizes the resin. Autoclaving, another sterilization method, can be done before packing the column.

After cleaning with base, the column bed should be re-equilibrated with starting buffer.

Superflow is an ideal support matrix for immobilizing biologically active substances. Superflow exhibits the very low nonspecific binding. This is most important when small amounts of biologics need to be isolated by affinity methods (e.g. diagnostic tests) or when a very high degree of purity is required (injectable therapeutics).

Uniflow 4

Uniflow 4 is an agarose-based gel filtration medium with bead-shaped particles, suitable for gel filtration and as an affinity chromatography support. Uniflow is available at three different agarose concentrations: 2%,4%, and 6%.

As agarose concentrations increase, matrix porosity decreases. The exclusion limit for globular proteins varies from 4,000kD (6% beads) to40,000kD (2% beads). This allows the separation of a wide range of molecules including proteins, polysaccharides, and nucleic acids.

Uniflow is a highly stable medium. The matrix features a unique polysaccharide composition that resists biological degradation. Additional crosslinking greatly enhances the mechanical and chemical stability of the matrix and eliminates the problem of biodegradation.

Lectin Resin

Concanavalin A Superflow 4

Concanavalin A (Con A) Superflow 4 is an affinity resin for the purification of glycoproteins, glycolipids, and polysaccharides containing terminal α-D-mannopyranosyl, α-D-glucopyranosyl sugars or sterically related residues. The resin is made by stably coupling purified Con A lectin to our highly crosslinked 4% agarose beads.

Con A Superflow 4 resin doesn’t show leaching in commonly used buffers and is stable between pH range 4-12. Con A Superflow 4 should be stored in buffers containing CaCl2 and MnCl2, as these ions are required for its activity. Con A Superflow 4 can be used to purify IgM, exosomes, and other membrane vesicles. Sterogene Con A Superflow 4 resin is very robust, after usage Con A Superflow resin can be easily regenerated and can be utilized multiple times without significant loss in yield.

GNA Superflow 4

Galanthus nivalis Agglutinin (GNA) Superflow 4 is an affinity resin used to purify glycoproteins containing mannose sugars. The resin is prepared by covalently immobilizing purified GNA lectin to our highly cross-linked 4% agarose beads. GNA Superflow 4 is a highly stable resin with no sign of leaching at pH 3-12 and in various common buffers.

The resin can be used multiple times without any loss in binding capacity. Many enveloped viruses including HIV and SARS-CoV-2 contain mannose residues in their surface glycoproteins, and thus our GNA Superflow 4 can be efficiently used in the purification of these viruses and respective surface glycoproteins for vaccine or structural studies.

GNA lectin is isolated from the bulbs of snowdrop (Galanthus nivalis) plants by affinity purification on mannose-agarose resin. GNA is a tetramer of identical subunits and has a molecular mass of 52 kDa. Unlike other mannose-binding lectins, GNA is not a metalloprotein and it doesn’t require Ca++ or Mn++ for its optimal binding activity. GNA shows a strong affinity for structures containing (α 1-3) mannose residues, it agglutinates rabbit erythrocytes but not human erythrocytes. In human serum, it binds to α2 macroglobulin and specifically binds murine IgM.

Jacalin Superflow 4

Jacalin is a lectin isolated from Jack-fruit seeds (Artocarpus integrifolia) and specifically binds to α-D-galactose sugar. It is a homotetrameric glycoprotein lectin with a molecular weight of 40 kDa and specifically binds to human IgA, but not IgG and IgM. Jacalin is widely used as a high-affinity ligand to purify or deplete IgA from serum and colostrum samples.

Sterogene’s Jacalin Superflow 4 is a stable, high binding capacity affinity resin (>3 mg of human IgA/ml resin) and is available in a small kit size or to fit any purification scale.

WGA Superflow 4

Wheat germ agglutinin (WGA) is a group of closely related isolectins present in wheat (Triticum sp) and has a strong affinity for terminal N-acetyl glucosamine and chitobiose. WGA binds preferentially to dimers or trimers of N-acetyl glucosamine or N-acetyl neuraminic acid.

N-acetyl D-glucosamine is a ubiquitous sugar modification in several proteins and lipids and thus binds to many proteins in plasma/serum, bacterial cell wall peptidoglycans, chitin, and various membrane proteins.

WGA is stably coupled to Sterogene’s Superflow 4 resin. After regeneration, the resin can be utilized multiple times.

Papain Actigel

Papain is a nonspecific cysteine protease, which cleaves IgG at the hinge region generating three fragments of ~ 50kDa (two Fabs and one Fc fragment). Antibodies digested with Papain enzyme cannot carry out agglutination, opsonization, precipitation, or lysis, but the Fab fragments are active and would bind and neutralize the respective antigens. Coupling of Papain with the Superflow resin increases the stability of papain enzyme, facilitates controlled digestion, and avoids papain contamination in the digested products. The Fabs can be further purified on the Protein A-Superflow column to get rid of the Fc fragments, and the flow-through will have pure and active Fabs.

Pepsin Actigel

Pepsin is a nonspecific endopeptidase with optimum activity between pH 2-7. Pepsin cleaves IgG at the hinge below the disulfide bonds on the Fc region generating one divalent F(ab’)2 and several small fragments of the Fc region. The coupling of pepsin to the Superflow resin increases its stability, provides better control on digestion of IgG, and greatly simplifies the purification steps of F(ab’)2 as the digested products are free from pepsin contamination.
